Researchers assess the binding behavior of SARS-CoV-2 virus – News-Medical.Net

The binding of a SARS-CoV-2 virus surface protein spike -- a projection from the spherical virus particle -- to the human cell surface protein ACE2 is the first step to infection that may lead to COVID-19 disease. Penn State researchers computationally assessed how changes to the virus spike makeup can affect binding with ACE2 and compared results to those of the original SARS-CoV virus (SARS).

The researchers' original manuscript preprint, made available online in March, was among the first to computationally investigate SARS-CoV-2's high affinity, or tendency to bind, with human ACE2. The paper was published online on Sept. 18 in the Computational and Structural Biotechnology Journal.

The work was conceived and led by Costas Maranas, Donald B. Broughton Professor in the Department of Chemical Engineering, and his former graduate student Ratul Chowdhury, who is currently a postdoctoral fellow at Harvard Medical School.

We were interested in answering two important questions. We wanted to first discern key structural changes that give COVID-19 a higher affinity towards human ACE2 proteins when compared with SARS, and then assess its potential affinity to livestock or other animal ACE2 proteins."

The researchers computationally modeled the attachment of SARS-CoV-2 protein spike to ACE2, which is located in the upper respiratory tract and serves as the entry point for other coronaviruses, including SARS. The team used a molecular modeling approach to compute the binding strength and interactions of the viral protein's attachment to ACE2.

The team found that the SARS-CoV-2 spike protein is highly optimized to bind with human ACE2. Simulations of viral attachment to homologous ACE2 proteins of bats, cattle, chickens, horses, felines and canines showed the highest affinity for bats and human ACE2, with lower values of affinity for cats, horses, dogs, cattle and chickens, according to Chowdhury.

"Beyond explaining the molecular mechanism of binding with ACE2, we also explored changes in the virus spike that could change its affinity with human ACE2," said Chowdhury, who earned his doctorate in chemical engineering at Penn State in fall 2019.

Understanding the binding behavior of the virus spike with ACE2 and the virus tolerance of these structural spike changes could inform future research on vaccine durability and the potential for the virus to spread to other species.

"The computational workflow that we have established should be able to handle other receptor binding-mediated entry mechanisms for other viruses that may arise in the future," Chowdhury said.

Glue is also expanding the library of ready and customizable spaces that teams can use with a new space called Mont Matiz a hill-top, open-air space that provides users with a relaxing setting for creative workshops and large events like employee town halls.

How Can You Be Sure Your Work Really Counts? – Forbes

Okay, you invest a lot of hours in your work. But is it producing the results you really want?

As the great philosopher Forrest Gump once postulated, Life is like a box of chocolates. You never know what youre gonna get.

That seems to be true with so many things. Ordering a meal at a new restaurant comes to mind. Or hiring someone to repair that gizmo you bought online. Or trying out the weight loss regimen thats incessantly pitched on that infomercial.

But with some things in life, mindful attention to certain human behaviors really can produce predictable results.

Take work, for example.

Theodore Roosevelt, himself one of the most active and productive people in our history, had this to say: Far and away the best prize that life has to offer is the chance to work hard at work worth doing.

Work worth doing. The definition is certainly in the eye of the beholder. But most of us would agree thatbecause the majority of our waking hours are invested in some kind of workensuring that it produces something good is worthy of attention.

Richard Lee shows the way. As one of Silicon Valleys top leadership trainers, hes coached senior teams at companies like Google, Facebook, Cisco Systems and Genetech/Roche.

In addition to being a gifted coach, Richard is a gifted listener and observer. He shares some of his key learnings in his new bookWORK THAT COUNTS: Breaking Down the Barriers to Extraordinary Results.

Rodger Dean Duncan:To help people produce high-impact work results, you use what you call the ACE Model (Alignment,Collaboration,Empowerment) to underscore three important mindsets. Why these particular three?

Richard Lee:These three issues came up more often than anything else in my decades of work with many of the worlds best innovation companies.

As organizations become more complex, it becomes easy for people to lose sight of the bigger picture. Theres a need for people to align around a common purpose and do the right thing, not just for their own immediate team, but for the broader organization.

People need to collaborate, not only within their own teams, butacrossteams. Most people know this already, but they find that partnering across teams often causes conflict within their teams.

This poses questions for team leaders: When is it right to let go? When to hold onto the reigns more tightly? Most managers lean too far in one of these directions. Finding the sweet spot is what leads to empowered relationships.

Duncan:You say these three mindsets are not only linked but are interdependent. Why is that important?

Richard Lee

Lee:Many organizations seem to deal with these topics in isolation. If any one of these three is weak, it will disproportionately diminish impact. If all are strong in and sync, they will drive inordinately greater impact.

I remember someone saying that if you arent aligned, you cant collaborate, so why try?I responded with But you cant get aligned without collaboration.And dont forget the fuel so people can take action and drive decisions. We need all three to make our work truly count.

Duncan:Empowerment is certainly an idea that appears in many leadership discussions. What do you see as some of the misconceptions about genuine empowerment, and what can leaders (and team members) do to ensure empowerment brings out the best in all parties?

Lee:Lets start with the misconception that its easy, or that an organization can somehow mandate it as a value and therefore a reality. Most companies mistakenly rely on their managers alone to build a culture of empowerment. I believe the way to building a better culture is dynamic.The team leader needs to let go and lift up, while team members must step up and earn it.Both parties contribute to either strengthening or diminishing the trust that drives this capability.

Duncan:What role does trust play in effective work teams, and what are some observable behaviors that help create and maintain trusting relationships?

Lee:Trust is key to effective, empowered relationships. A key skill for all team members is credibility buildingthe ability to set expectations and meet them, adjust expectations as necessary, and knowing when to ask for help.

If team leaders believe their team members will ask for help when its needed, they will be more confident that the team members are less likely to overextend themselves and create unnecessary risk.

Talking it out candidly and constructively, along with investing in those relationships by sharing information and genuinely caring about those in their ecosystems, are also key to building trust.

Duncan:Interdependence is of course a key ingredient of effective teams. What do you see as best practices in managing mutual expectations?

Lee:People tend to think oftheirteam as the team their manager leads. But the truth is that your real team is bigger than you think. Think of the larger enterprise, or at a minimum, the larger division of which your department is only a part. Explicitly identify interdependencies while defining quarterly goals. And consider what you need from other groups and what other groups are depending on from yours.

One of the skills I explore on alignment is how to make the most of each meeting by clarifying the what, the why, and the how. The why gets at mutual expectations. Why is this project, this request important? How does it fit in the bigger picture?

Duncan:Moving decision-making downward is necessary for an organization to scale and grow. How can leadersat all levelsdo this most effectively?

Lee:The first step is for leaders to fully comprehend the risks of concentrating all decisions at the top. Its also important to recognize that in complex organizations where so much work is interconnected across teams, the leader usually cannot delegate full decision making unless its relatively contained and narrowed. This is why I refer more todrivingdecisions rather thanmakingdecisions.

It helps to realize that the decision-making process is dynamic. Of course, focusing on less risky decisions can make this more palatable. Ask your team members in which areas they want to drive decisions, and how they can include you along the way.

Duncan:Accountability is often associated with punishment and other negative connotations. How can leaders position accountability in a helpful framework and use it to encourage people in producing great work results?

Lee:As we all know, actions speak much louder than words.The manager needs to show over time through their behavior that their intention is to fix issues rather than focus on blame.

The leader should call it out if someone misses a commitment or behaves unacceptably, and do so in a relatively neutral manner. Then move to exploring together, as two adults, what can be done to mitigate the consequences or fix the problem. Remember that these moments are learning opportunities, so focus on what each party can learn.

Steve Wozniak-backed token WOZX skyrockets ahead of second listing – Cointelegraph

Apple co-founder Steve Wozniaks new token, Efforce (WOZX), almost doubled in price today after an astonishing run that saw the price increase around 26 times in the past week.

WOZX will open for trading on South Korea-based exchange Bithumb on Dec. 9, and is already up 2,490% since first being listed on the HBTC exchange on Dec. 3. It opened at $0.10 per token and is currently trading at $2.59.

Ethereum blockchain token WOZX is a way to securitize energy savings. Thetoken has a total supply of one billion and was sold via private funding rounds conducted earlier this year, receiving an initial valuation of $80 million.

It reached a market cap of $950 million in its first 13 minutes of trading, according to the eponymous company behind the token, Efforce.

It is Wozniaks second blockchain-related venture since co-founding Apple in his parents garage in 1976. Touting itself as the first blockchain-based energy saving platform, Efforce aims to encourage the reduction of energy consumption in a way that is meaningful but does not disrupt current human behavior or routine.

WOZX tokens will be used on the platform by contributors who want to take part in energy-saving projects and as rewards based on the amount of energy a user has saved.

Wozniak is joined at Efforce by co-founders Jacopo Visetti and Jacopo Vanetti, who serve as project lead and chief technical officer, respectively. The company was founded in 2019 and is based in Malta, a nation widely considered to be friendly to blockchain-based enterprises.

In a Dec. 4 announcement, Visetti described how Efforce will help democratize the energy efficiency market by connecting investors with energy savings projects.

In Oct. 2018, Wozniak founded the blockchain-based capital venture fund EQUI Global in an effort to disrupt the venture capital and funding industry.

Anatomy of a Goal: Artur books the Columbus Crews ticket to MLS Cup – Massive Report

Welcome back to the Anatomy of a Goal, where each week we dissect one goal (or near goal) from the Columbus Crews previous match.

For the 2020 MLS Eastern Conference Final, we take a look at Arturs 59th minute goal that gave the Crew a 1-0 win against the New England Revolution, setting up Columbus as the host of the 2020 MLS Cup.

Here is a look at the historic goal from the Black & Golds Brazilian midfielder.

The Crew took the field against New England knowing that a win would bring the MLS Cup Final to Columbus the following week. First, the Black & Gold would have to silence the Revolutions potent attack.

The first half of the 2020 Eastern Conference Final went almost exactly as the Crew dreamt despite the 0-0 score at the whistle. Columbus controlled 54 percent of possession and put five shots on goal to the Revolutions zero. But for some excellent goaltending by Matt Turner, the Black & Gold would have entered halftime with at least one goal.

Arturs game-winner begins with a Crew corner kick in the 59th minute. Lucas Zelarayan lines up to hit an in-swinging kick into the goal box .

As Zelarayan steps up to the kick, Columbus attackers begin their movements. Darlington Nagbe (6 in the above picture) situates himself at the top of the 18-yard box to collect any ball that makes its way out and to help break up any Revolution counter attacks. Gyasi Zardes (11) is positioned on the edge of the six-yard box to flick any short corners into the front of the goal. Artur (8, to the left of Zardes) is set up in a similar spot and is tasked with heading the ball directly on goal. Jonathan Mensah (4) and Josh Williams (3) are the two direct headed-shot targets in this play. Mensah roams free in the middle of the penalty box while Williams will collect any ball that reaches the back post.

Zelarayans corner makes its way to Mensah who is marked by Scott Caldwell.

The kick just misses Matt Polster but neither Mensah nor Caldwell are able to get a clean touch onto the ball.

Caldwell ends up on the ground while the both teams search for the ball, which just happens to have fallen right in to the side of Polster.

Polster finds the ball and quickly must decide whether to clear the ball, play a quick pass forward to Gustavo Bou (7) or make a short pass to Tajon Buchanan (17).

Polster scuffs an attempted clearance but slices the ball right to Buchanan.

Buchanan keeps his eyes on the ball and tees up a clearance.

The Revs young right back is able to get off his clearance and sends the ball toward midfield.

Harrison Afful, one of the shorter players for the Black & Gold, stayed back on defense during the corner kick and is in position to pick up Buchanans clearance as the ball takes an awkward bounce just before reaching him.

Afful has time to settle the ball but hits an awkward attempted pass back into the 18-yard box after the ball takes a difficult bounce.

Affuls attempted pass ends up soaring high into the air and falling down between Pedro Santos and Luis Diaz. The Crew attackers battle Carles Gil for possession of the ball.

Diaz out-jumps Gil and glances a headed pass to Zelarayan.

Zelarayan easily receives Diazs header and Gil shits his defensive attention to Columbus No. 10.

Zelarayan collects the ball and the Black & Gold begin a secondary attack. Mensah had tracked back defensively, but sees open space ahead of him. Williams tracks back to provide defensive cover as his center back partner rejoins the attack. Artur also shifts forward into an attacking position.

Zelarayans deft ball-work to beat Gil and send a cross into the penalty box is worth a look at in slow motion before we break it down.

First, Zelarayan makes a hard cut with his left foot, sending the ball a few feet toward midfield and setting Gil off balance. Artur and Mensah continue their runs toward the goal.

Gil scrambles to recover while Zelarayan prepares a quick Cruyff turn where he plants his left foot, sends the ball behind his left leg with his right foot and speeds off the other direction.

From this angle, you can see just how much open space Mensah has ahead of him.

Zelarayans Cruyff turn sets Gil off balance and sends the Crew attacker toward the goal.

From the TV angle, you can see Gil plant hard on both feet after Zelarayan has already gone the other direction.

Zelarayan carries forward with Gil trailing. Artur and Mensah set up at the top of the 18-yard box. From this angle, you can see Artur and Mensah turn to look at each other. Both players know exactly where the other will be, setting up Mensahs eventual pass into the path of Artur.

Zelarayan is out in front of Gil and has four options. He can play a quick pass forward to Santos, attempt to carry the ball forward, try a difficult through pass to Zardes or a back-post cross to Mensah.

Columbus No. 10 spots his captain at the back post and hits a long cross into the 18-yard box.

Mensah settles into position. If the ball makes it past Buchanan, the captain will have the chance to take a first-touch shot or make a quick pass. Artur sprints into the box to provide a passing option for his teammate.

Mensah is too far out to try a headed shot so he prepares to receive the ball with his feet. Buchanan is about three yards away and just turns in time to see Mensah prepare to receive the ball. Artur sprints, unmarked, toward the top of the box.

Mensah receives the ball and can either take a touch toward the goal, hit a first-touch shot on goal play a quick pass into the path of Artur or play a pass toward Zardes on the other post.

Mensah knows that Artur is trailing just behind him and that he will be making a run into the box, so he plays a first-touch pass toward the midfielder.

Polster is on the wrong side of Artur, giving the Brazilian a direct path toward the ball.

Artur approaches the ball, with Polster attempting to defend with four options. He can try to find Zardes near the six-yard box, hit a first-touch shot on goal, attempt to carry the ball forward or play a pass back to Mensah.

Artur hits a first touch shot with his left foot. Somehow, the Brazilian is able to put a slight curve on the ball that sends it just inside the goal post.

Arturs shot speeds between Henry Kessler and Buchanan leaving Turner as the Revolutions last line of defense.

Turner lays out for the ball as it rolls toward the goal line.

The ball misses Turners finger tips by a few inches . . .

. . . beating the New England goalkeeper and rolling . . .

. . . into the back of the net!

Anatomy of a Play: Justin Herbert has his mind read again – Touchdown Wire

Los Angeles Chargers rookie quarterback Justin Herbert has enjoyed a strong rookie campaign on a personal level. Herbert stepped into the starting lineup under rather interesting circumstances, but during his time under center the rookie has demonstrated pocket, athleticism and arm talent beyond what most expected from him coming out of Oregon.

To date, Herbert has completed 64.8% of his passes for 3,224 yards and 23 touchdowns, against just nine interceptions. Very solid numbers for a rookie passer on a team that has a 3-9 record.

Now as we start to think about his future, his development path and ways he can improve over the final few games of 2020 and into 2021, there is an area to watch. As we illustrated last week in a breakdown of TreDavious Whites interception, Herbert has made mistakes when making assumptions about the backside defender in zone coverage schemes.

That happened again on one of his two interceptions against the New England Patriots. Prior to Chase Winovich stepping in front of a crossing route Herbert had almost all the information he needed to make a big play, except for one piece: Where Winovich was on the field.

Lets turn to the tape:

Herbert has shown tremendous potential as a rookie. As we think about his next season, this is an area to watch for development. If he can start seeing the full picture when making decisions, he will be an even more difficult quarterback to defend.

Anatomy of a Bust: How 49ers miscommunication led to an easy Bills touchdown – Touchdown Wire

With 9:50 left in the Bills Monday night game over the 49ers, and Buffalo already up 27-17, quarterback Josh Allen threw a 28-yard touchdown pass to rookie receiver Gabriel Davis in which several San Francisco defensive backs appeared to be very confused. It was a bad play for the 49ers in what turned out to be a 34-24 win for Buffalo.

Pre-snap, you can see cornerback Richard Sherman communicating with safety Dontae Johnson. What might Sherman have been saying? Sherman was probably imploring Johnson to make an alert call to run the coverage to the actions of slot receiver Cole Beasley. Beasley ran a quick five-yard in-cut, while Davis ran up the boundary. In a basic Cover-1 or Cover-3 call, you probably have Sherman on Davis to the boundary, but based on Beasleys actions, that was never the call. Sherman expected to take Beasley inside, while somebody playing safety (perhaps Dontae Johnson) was supposed to cross and take Davis to the outside. Its a coverage called Palms, and it was an unfortunate bust for Robert Salehs defense. Safety Tarvarius Moore was also in the deep third to that side, so yikes.

We were in Palms coverage, Sherman explained after the game. Two into the flat. I adjusted, but we had a few busts on the play. Its unfortunate. The motion put us in a look we hadnt seen before. Its just one of those plays just a miscommunication down the line, and you cant have those plays in games like this.

The look the 49ers hadnt seen before may have been Beasley going in motion from left to right, which could have confused the coverage rules. Shermans alert call should have taken the coverage to that side from zone to Palms.

For those who dont know, Palms coverage can also be called 2-Trap or 2-Read or Soft-2, based on which playbook its in. When I watched tape with Sherman in 2015, he explained how the Seahawks used Palms coverage against Dez Bryant and the Cowboys on this play in which Sherman was playing outside until Bryant took the quick screen behind the line of scrimmage. At that point, Sherman was supposed to crash down on Bryant, which he did, for a three-yard loss. It appeared as if there was a coverage void over Bryant in the slot with Sherman outside, but thats exactly how it was supposed to look.

Its like they call it Soft 2, Sherman told me back then. You have two receivers to your side. If 2 [the slot receiver] goes to the flat five yards or less, the cornerback takes him. If 2 goes vertical past five yards, the corner has man coverage on 1 [the outside receiver]. Since he went bubble [ran a bubble screen], thats five yards or less, and you shoot your gun. Im on 2 until he gets past this point, and then Im on 1. Which sounds easy until I still had to take a gamble on that play, because if I go underneath and I miss, [Bryants] got at least 2030 yards. Then, [safety] Earl [Thomas] will have to chase him downitd be a footrace between him and Dez. If I take that and [Bryant] blocks me, hes up the seam.

So, if Bryant had gone up the seam, the safety would have been responsible for him, and Sherman would have stayed on the outside receiver. The defining point is the slot vertical (if its there) after five yards. Holding the safety back might give a bust disguise look pre-snap. Of course, it helps if your safety is Earl Thomas in his prime.

As this X&O Labs article shows, the responsibility for the alert call into Palms could come from the playside safety in certain instances. We dont know exactly how the miscommunication started in this case, but we sure know how it ended with one of the easiest touchdowns Josh Allen will ever experience.

The review was conducted by an age group of women who underwent assisted reproduction and in relation to the method applied to achieve pregnancy.

A special standard was developed for the compliance review procedures in accordance with European Union guidelines on criteria to ensure qualitative and objective statistical reporting, international standards for data verification and the principles of the European Statistics Code of Practice and European Commission Regulation 2017/0048 (COD).

Ms Angeliki Katsapi, Executive Director of the EuroMediterranean Institute for Quality and Safety in Health Services and of Temos Hellas, said: "The verification of the Unit's clinical results focuses on the origin of data and the way they are used in publishing statistical reports and, specifically, to the evaluation of their accuracy, transparency, consistency and comparability. I believe that this innovative certification of clinical results should serve as the beginning of more broadly applied certifications in the sector so that every woman, every couple, are able to obtain full and objective knowledge of the services being offered by health providers."
